Wheat gluten proteolysis by enzyme preparations of directional action

چکیده
A comparative study of enzymatic hydrolysis of dry wheat gluten was carried out by exoprotease and endoprotease enzyme praparations. Gluten of different rheological properties was conditionally divided into three groups on the basis of the compressive deformation index (Hdef) determined with the IDK-1 instrument: shortcleaved (Hdef 40 instr. units), good (Hdef 65 instr. units) and weak gluten (Hdef 80 instr. units). The optimal conditions for hydrolysis of different quality gluten by endoprotease enzyme praparations (Neutrase® 1.5MG, Protamex®) and exoprotease enzyme preparation (Flavourzyme® 500MG) were determined. The degree of proteolysis was assessed by the number of released amine nitrogen. It was shown that the stronger gluten is, the greater amount of amine nitrogen is formed under the influence of endoprotease (Neutrase® 1.5 MG, Protamex®), the weaker it is, the more nitrogen is formed under the influence of exoprotease (Flavourzyme® 500 MG). The most effective parameters of hydrolysis (time, concentration of the enzyme preparation, temperature, pH) revealed by us were specified by electrophoresis in polyacrylamide gel. The degree of hydrolysis necessary for achieving a certain molecular mass of peptides when used in breadmaking was determined. Thus, a process of hydrolysis of dry wheat gluten, aimed at expanding the range of its uses for improving and enriching wheat bread, was developed. We established that the functional properties of dry wheat gluten can be regulated by the change of the enzymatic hydrolysis time in the presence of the enzyme preparation Protamex®. * Corresponding Author: V. Improving the rheological properties of 18% wheat flour as affected by transglutaminase enzyme
Enzymes are useful to modify wheat proteins to preserve the gas better and to correct the rheological properties of the dough of weak flour and bread. Gluten proteins are highly impacting the quality of various gluten-based products, and transglutaminases (TGs) leading to the strengthening, stability and constancy of the dough as well as the improvement of the volume, texture and storage time o...
متن کاملStudies on the Mechanism of Destruction of the Toxic Action of Wheat Gluten in Coeliac Disease by Crude Papain.
The toxic action of wheat gluten on two patients was eliminated after predigestion of gluten by crude papain. This change is thought to be due to an enzyme which liberates ammonia from gluten. It is not thought that this particular mechanism operates in the normal intestinal cell but it is envisaged that one of the peptide bonds of the coeliac active constituent (possibly a N-glutaminyl peptide...
متن کاملInteractions of Hydrocolloid and Emulsifier on Rebuilding Gluten Network of Damaged Wheat and Taftoon Bread Quality
Many factors can affect wheat gluten properties and functionality. One of the most important, of these factors, is wheat bug damages. Insect injects a salivary proteinase into wheat kernel that results a significant weakening effect on the gluten, in substantial losses in physical quality, dough properties and baking performance. Bread baked with the insect – damaged flour had poor quality and ...
متن کاملPartial Purification of Deamidase from Germinating Wheat (triticum Aestivum L.) Seeds and Its Application for the Improvement of Functional Properties of Wheat Gluten
Enzyme with deamidase activity was found in the germinating wheat seeds. The activity appeared in the seeds after 24 hours of germination and reached its maximum value after 48 hours and then declined rapidly. Protein deamidase from germinating wheat seeds was further purified by salting out with phosphate salts and by subsequent chromatography on gel filtration and DEAE-cellulose column chroma...

Wafles In the production of biscuits and waffles, gluten-weak flours are normally required. Previously there was no difficulty in obtaining such flours in Europe, but as more and more strong gluten varieties of wheat are grown in every country, this has become an area of some concern. High-gluten varieties of wheat intrinsically bind more water and this means that to achieve a lower viscosity, ...
